Gloom on the Amiga CD32 has a version of Defender in it.
Not sure about the normal Amiga version?

Playing Galaga while loading Tekken + complete the 8 levels you get Devil Kazuya!

If do the following while the game is loading hold Up + L1 + Triangle + X on controller 2 you can get 2-player Galaga!

Yep that's correct.

If we're talking games included as seperate bonuses I'm pretty sure Metroid Prime and Metroid Zero Mission contain the original NES Metroid to unlock. Think you might need a GBA and Metroid Fusion to ulock it via Metroid Prime though.

For genuine games within actual gameplay Timesplitters 2 has 3 retro style games to find and play during the story mode via the temporal uplink.

Maniac Mansion in playable on the PC you find in the sequel Day of the Tentacle.

Lotus 2 on the Amiga has a hidden shooting gallery game you can access with a code.

The Project Gotham games, of course, have Geometry Wars.

There's a playable arcade game in GTAIV isn't there? Or am I making that up?

Also, the SNES version of Prince of Persia is hidden in The Forgotten Sands on the Wii. The 360 and PS3 versions, IIRC, contain PoP2 instead.

It's Rogue Squadron III: Rebel Strike, it contains both Star Wars and The Empire Strikes Back vector arcade games.

Starblade (Alpha) is hidden away as un unlockable in Tekken 5 on the PS2
WANTED: Mario Party 3 on N64 (just the Box only) ANY SNES GAMES, OR EMPTY SNES BOXES, INSTRUCTION MANUALS or WHITE INNER TRAYS.
