It's a tough choice between The Way of the Exploding Fist (C64), Barbarian (Amiga), and Body Blows Galactic (Amiga).

I'll go with the superlative Super Street Fighter 2 Turbo. The best of the Street Fighter 2 series, which is still played in pro tournaments today as a testament to its longevity.

Surely something on the Neo-Geo, you know, the fighting game console?! I vote for Garou: Mark of the Wolves or Real Bout Fatal Fury Special.

For me it's a toss up between Soul Calibur & Street Fighter Alpha/Zero 3.
The square is a great shape, and one that is criminally underused in modern game design. In the push to ever more granular, chaotic environments, games have lost sight of the simple pleasures of right angles and straight lines.

The 32x version of Virtual Fighter. Plays brilliantly, and an amazing conversion as well. Far superior to the Saturn version.

Barbarian on the C64, as I child nothing was cooler then beheading an opponent and seeing the severed head kick across the screen by a goblin.

International Karate on the Commodore 64.
Simplicity is key. It took Way of the Exploding Fist and improved on it in every way, with better graphics, sound and most importantly, gameplay and AI. It doesn't over-complicate things with odd joystick combinations... you simply choose your move and execute it with a move of the joystick and, if necessary, a press of the button.
That's all you need.
The perfectly-judged soundtrack adds to proceedings, kicking up a notch just as the opponents get more difficult, making every fight seem even more frantic. You never lose because the opponent pulls off a cheap move... you lose because you make a mistake.
IK+ was not as good a game, for me. The addition of an extra fighter took away from the purity of the original. I never really got on with fighting games that require insane joystick combinations to pull off special moves. To me, nothing was quite as effective as a well-delivered smack to the face. And for that, International Karate on the C64 remains king.
I will throw in a special mention for the arcade version of Yie Ar Kung-Fu, though. It introduced the world to the concept of opponents with different looks, styles and even weapons. Also, I'm really good at it.

From the Psone era I really enjoyed the Tobal and Rival Schools games. Not so widely known but none the less enjoyable.

Barbarian on the CPC, there has never been another fighter like it since.

I'd say probably Street Fighter Alpha 3 Upper, I'm no good at fighters at all and it's one of the very few fighters that I have actually 1CCed.
